Revealing predictive proteomic insights

Robust immunophenotyping strategies for drug discovery and development are crucial to better understand the immune response to disease and therapeutics. The challenge is capturing a wide enough range of cell types and their functions to account for biological heterogeneity –essential for identifying predictive biomarkers, stratifying patients and understanding mechanisms of action. High-dimensional panels comprised of intracellular and surface markers are ideal for clinical cohorts, as they increase the amount of information acquired from limited precious samples. Representative dot plots show the compatibility of the Human Broad Immune Profiling CyTOF Panel, 20 Antibodies with stabilization reagents and sample multiplexing in three different workflows.

CyTOF technology has exceptionally high reproducibility, as metal-tagged antibodies can withstand the harsh elements of time and temperature. In a comparison study with different experimental workflows, CyTOF technology demonstrates reproducible data quality. For all 32 immune cell populations ≥5% in frequency, the %CV within each donor across the different workflows was <9.2%. In addition, frozen antibody cocktails are stable and yield comparable signal intensities over time.

An easy-to-implement solution for routine and reproducible immune profiling

This video highlights a streamlined and standardized workflow for the Maxpar™ Direct Immune Profiling Assay, a dry-format 30-marker antibody panel optimized for deep phenotyping of human peripheral whole blood and PBMC. This robust assay combines a single-tube workflow with an automated five-minute data analysis solution to quickly phenotype 37 immune cell populations, with no titrations needed.  It provides researchers with a routine and reproducible method to better understand the variation in immune response to disease pathogenesis and therapeutic interventions.

Technology

Cytometry by time-of-flight or mass cytometry (on which CyTOF technology is based) is a proteomic technology that uses metal-conjugated antibodies to simultaneously analyze 50-plus intracellular and surface markers in biological samples at single-cell resolution. This overcomes the limitations of spectral overlap and post-stain sample instability associated with fluorescence flow cytometry, enabling greater immune coverage, higher reproducibility and consistency, and faster execution of large immunophenotyping studies.
